Now that I have used the new cards for a few days, I can say that the speed increase is quite noticeable even during normal use. I say this because I don't "feel" the lag of uncompressed Nef's like I did before. But the real surprise was the downloading speed of the raw files to my computer.
I have a built-in card reader connected to a USB 2.0 port and the difference in downloading compared when using the same reader with the Extreme III cards is phenomenal. It really is double the speed if not more!
Still, my recommendation is that the extreme III cards are fast and will do well in a D300, but if you can get a good trade-in price go for the upgrade, the faster downloading from card to PC and the faster downloading from camera to card are nice to have and help make the experience with the camera even better.
